Changing link titles

hard to say if there is a penalty

no problem if new keywords in url are contained in the body of the page imo. I would also use a 301 redirect from old url to new url.

Are you refering to “anchor text” (like in my sig where it says “Microgaming Casinos”)

or the actual url?

you can change anchor text all you want without penalty.

Assuming you are talking about changing a URL. If you change the URL, you need to 301 redirect it to let Google and others know that you have moved the page. It will take a few weeks, but whatever rankings and SEO benefits you had for the old page will be moved over to the new page.
